Quick question about amp gain

Hello,

I'm a bit confused seeing as there is a way to set amp gain with multimeter right.

Setting it with multimeter you would get your target voltage by doing √(Output amp RMS x Ohms).

But isn't the point of amp gain is to match it to your headunit output voltage? How does the calculations made to find target voltage relate to the headunit output voltage?
